SULFTRIM 480 utilizes a highly effective sequential blockade mechanism to achieve powerful bactericidal synergy. Sulphadiazine acts as a competitive inhibitor of PABA, disrupting the synthesis of dihydrofolic acid (Stage 1). Trimethoprim then blocks the subsequent enzyme, dihydrofolate reductase, preventing the production of tetrahydrofolic acid (Stage 2). Since both steps are essential for bacterial DNA and RNA synthesis, this dual blockade kills the bacteria more efficiently than either agent alone, preventing resistance and ensuring broad-spectrum activity.
